Description
Intestinal alkaline phosphatase 1 plays a central role in gut barrier defense and lipopolysaccharide detoxification, making precise enzymatic characterization essential for understanding its signaling functions. This recombinant rat Alpi, covering the full mature protein (aa 21–511) with an N-terminal 10×His tag positioned away from the catalytic domain, demonstrates a specific activity exceeding 10,370 U/mg against pNPP at 37°C and pH 10.0—a level that supports rigorous kinetic parameter analysis (Km, Vmax, kcat/Km), substrate specificity profiling, and inhibitor screening with reliable IC50 determination. Expressed in mammalian cells to preserve native folding and post-translational modifications, this enzyme is also appropriate for use as a positive control in enzyme-linked assays and for drug candidate evaluation targeting alkaline phosphatase inhibition mechanisms. Purity greater than 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for accurate kinetic measurements and structural studies such as crystallography.
