Alkaline phosphatase, germ cell type plays a critical role in embryonic development and germ cell differentiation, yet its catalytic properties have been challenging to characterize due to limited availability of active recombinant material. This full-length mature protein (aa 19–502) demonstrates robust enzymatic function with a specific activity exceeding 6000 U/mg against p-nitrophenylphosphate at pH 10.0, providing quantitative performance data that supports kinetic parameter analysis including Km and Vmax determination, as well as inhibitor screening with reliable IC50 measurements. The C-terminal 10xHis tag preserves native catalytic site architecture, enabling accurate substrate specificity profiling and mechanism-based inhibition studies without steric interference at the active site. Purity greater than 95% by SDS-PAGE and endotoxin levels below 1.0 EU/μg meet the quality thresholds commonly required for enzyme-linked assays where this protein serves as a positive control, and satisfy criteria typical for structural biology applications including X-ray crystallography.
