Description
Semaphorin-4D plays a pivotal role in immune cell regulation, influencing T-cell activation, B-cell responses, and tumor microenvironment remodeling through interactions with plexin receptors and CD72. This recombinant human SEMA4D fragment (aa 22–734) carries a C-terminal 10×His tag and is produced in mammalian cells to preserve native-like glycosylation and proper folding of the sema domain. Functional ELISA confirms robust binding to Pepinemab, with an EC50 of 4.035–5.743 ng/mL when immobilized at 2 μg/mL, providing a suitable basis for anti-SEMA4D antibody screening, receptor-ligand binding assays, and development of blocking-antibody characterization platforms. Purity exceeding 90% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based functional assays investigating SEMA4D-mediated immune signaling and for use as a reliable reference standard in quantitative immunoassays.
