Description

PD-L1 is a critical immune checkpoint ligand whose therapeutic targeting demands conformationally accurate recombinant protein for reliable antibody screening and inhibitor discovery. This human CD274 construct spans the extracellular IgV and IgC domains (aa 19–238) and carries a C-terminal hFc1 tag, enabling oriented immobilization in binding assays while mammalian cell expression preserves native glycosylation and disulfide-dependent folding. Functional ELISA confirms robust activity, with an EC50 of 1.252–1.653 ng/mL against an anti-PD-L1 monoclonal antibody, supporting use in competitive blocking antibody screening, epitope mapping, affinity characterization by SPR or BLI, and small-molecule checkpoint inhibitor discovery. Purity exceeding 95% by SDS-PAGE combined with endotoxin levels below 1.0 EU/μg satisfies the criteria typical for cell-based immune checkpoint co-culture assays and sensitive receptor-ligand interaction studies.

Applications : Enzymatic activity in vitro

Review: I used the CSB-MP878942HU1 protein for an ELISA experiment, and the EC50 was measured to be 0.3038-0.6364 ng/ml. The results showed that the protein has good biological activity and can be directly used for subsequent drug-related research.

As a ligand for the inhibitory receptor PDCD1/PD-1, modulates the activation threshold of T-cells and limits T-cell effector response. Through a yet unknown activating receptor, may costimulate T-cell subsets that predominantly produce interleukin-10 (IL10).; The PDCD1-mediated inhibitory pathway is exploited by tumors to attenuate anti-tumor immunity and escape destruction by the immune system, thereby facilitating tumor survival. The interaction with PDCD1/PD-1 inhibits cytotoxic T lymphocytes (CTLs) effector function. The blockage of the PDCD1-mediated pathway results in the reversal of the exhausted T-cell phenotype and the normalization of the anti-tumor response, providing a rationale for cancer immunotherapy.
