Description
GUCY2C's extracellular domain has emerged as a high-value target for therapeutic antibody development in gastrointestinal oncology, and this construct spans residues 24–430 to capture the complete ligand-binding region critical for receptor-antibody interaction studies. Expressed in mammalian cells to preserve native glycosylation and conformational integrity, the C-terminal hFc1 tag facilitates oriented immobilization in binding assays—validated by functional ELISA demonstrating an EC50 of 3.049–4.660 ng/mL against an anti-GUCY2C recombinant antibody at 5 μg/mL coating concentration. This quantitative binding performance supports use in competitive inhibition assays, blocking antibody screening, therapeutic antibody epitope mapping, and affinity characterization by SPR or BLI. The protein satisfies the purity and endotoxin criteria typical for sensitive functional assays, with greater than 90% purity by SDS-PAGE and endotoxin levels below 1.0 EU/μg, providing a suitable basis for drug discovery screening of biologics targeting GUCY2C.
