Description
CD276 (B7-H3) functions as an immune checkpoint molecule with context-dependent co-stimulatory or co-inhibitory roles in T-cell activation, making it a target of interest in cancer immunotherapy research. This mammalian-expressed construct spanning residues 29–245 of isoform 2 captures the first two immunoglobulin-like domains, preserving the native glycosylation and folding required for authentic receptor-antibody interactions. Functional ELISA validation demonstrates specific binding to an anti-CD276 recombinant antibody with an EC50 of 47.12–54.16 ng/mL, providing quantitative evidence that this protein supports therapeutic antibody epitope mapping, competitive inhibition screening, and blocking antibody validation assays. The combination of >95% purity and <1.0 EU/μg endotoxin meets the quality thresholds commonly required for surface plasmon resonance affinity characterization, biolayer interferometry kinetic studies, and use as a positive control in immune checkpoint binding assays.
