ACKR1 (Duffy antigen/chemokine receptor) is a seven-transmembrane receptor that presents unique challenges for recombinant production due to its multi-pass topology, and this full-length construct (aa 1–336) leverages a cell-free E. coli expression system to deliver the complete sequence—including all extracellular and transmembrane domains—without the truncation often necessitated by cell-based membrane protein expression. Functional ELISA confirms ligand-binding competence: immobilized ACKR1 at 1 μg/mL binds human CCL2 with an EC50 of 48.64–60.24 μg/mL, providing validated evidence that supports use in receptor-ligand interaction assays, competitive inhibition studies, blocking antibody screening, and therapeutic antibody epitope mapping. The N-terminal 10×His tag enables straightforward oriented capture on Ni-NTA biosensor surfaces for SPR or BLI affinity characterization of chemokine interactions or small-molecule inhibitor candidates. Purity exceeding 85% by SDS-PAGE aligns with standards expected in binding assay positive controls and early-stage drug discovery workflows targeting chemokine-mediated cardiovascular signaling.
