Description

Persephin signals through the GFRα4/RET receptor complex to promote neuronal survival, yet its recombinant production demands careful attention to the disulfide-rich cystine knot fold. This tag-free human PSPN fragment (aa 61–156) encompasses the mature, receptor-binding domain and demonstrates an ED50 below 10 ng/mL in TT medullary thyroid cancer cell proliferation assays, corresponding to a specific activity exceeding 1.0 × 10⁵ IU/mg. Such potency supports use in neuronal survival and proliferation assays, RET-dependent signaling studies, and receptor-ligand binding experiments via SPR or ELISA. Produced in *E. coli*—appropriate for this non-glycosylated growth factor—the protein meets purity greater than 97% by SDS-PAGE with endotoxin levels below 1.0 EU/μg, satisfying the quality criteria typical for cell-based bioassays and in vivo regenerative neuroscience models.
