LRP6

LRP6 encodes Low-Density Lipoprotein Receptor-Related Protein 6, a type I transmembrane protein also known as Arrow homolog. As a core co-receptor in Wnt signaling, it forms complexes with Frizzled receptors upon Wnt ligand binding, triggering β-catenin stabilization and nuclear translocation to regulate target gene transcription, critical for embryonic development and tissue homeostasis.
Dysregulated LRP6 is linked to osteoporosis, metabolic syndrome, and cancers like colorectal carcinoma. Its extracellular domain mutations impair Wnt signaling, while intracellular variants may enhance β-catenin activity. Therapeutically, monoclonal antibodies targeting LRP6 are explored for cancer, and small-molecule modulators are being developed to fine-tune its signaling in metabolic disorders.
