The protein encoded by the KLK1 gene, kallikrein-1, also termed tissue kallikrein or renal kallikrein, is a serine protease that cleaves low-molecular-weight kininogen to generate lysyl-bradykinin (kallidin). This bioactive peptide activates bradykinin B2 receptors, modulating vasodilation, inflammation, and sodium excretion. KLK1 operates within the kallikrein-kinin system (KKS), intersecting with the renin-angiotensin-aldosterone system (RAAS) to regulate blood pressure and renal function. It also interacts with mitogen-activated protein kinase (MAPK) and nitric oxide synthase (NOS) pathways, influencing endothelial homeostasis. Dysregulation of KLK1 has been implicated in hypertension, chronic kidney disease, and diabetic nephropathy due to altered kinin-mediated vascular tone and fibrosis.
Pharmacologically, recombinant KLK1 (e.g., human urinary kallidinogenase) has been evaluated in clinical trials for ischemic stroke and hypertension, demonstrating potential in improving cerebral perfusion and reducing endothelial dysfunction. Small-molecule agonists targeting the KKS pathway, including KLK1 activators, are under investigation to circumvent the rapid degradation of kinins. However, challenges persist in achieving tissue-specific delivery and minimizing off-target inflammatory effects, necessitating advanced peptide engineering or nanoparticle-based delivery systems to enhance therapeutic efficacy.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunofluorescence staining of Hela cells with CSB-PA012446LA01HU at 1:100, counter-stained with DAPI. The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. The secondary antibody was Alexa Fluor 488-congugated AffiniPure Goat Anti-Rabbit IgG(H+L).
