Description
Interleukin-4 (IL4) orchestrates Th2 differentiation and B-cell class switching, making precise receptor engagement essential for dissecting these pathways without confounding inflammatory signals. This recombinant protein binds human IL-4Rα with an EC50 of 7.9–9.8 ng/mL in functional ELISA and demonstrates a 17.1 nM affinity constant by surface plasmon resonance, providing quantitative benchmarks that support dose-response studies in T-cell polarization assays, B-cell proliferation experiments with primary PBMCs, and JAK1/STAT6 phosphorylation kinetics. Spanning the complete mature sequence (aa 25–153) and produced in mammalian cells to preserve native glycosylation, the protein meets the purity and endotoxin criteria typical for cell-based functional assays where lipopolysaccharide contamination above 1.0 EU/μg would trigger artifactual TLR4 activation and obscure IL-4–specific responses. The validated receptor-binding profile also makes this reagent appropriate for antibody development workflows, serving as a coating antigen in ELISA or as a positive control in neutralization assays.
