Human Complement fragment 5a,C5a ELISA Kit

The human C5a ELISA kit is a solid-phase immunoassay specially designed to quantitatively measure human C5a in serum, plasma, or tissue homogenates. It is based on the Sandwich-ELISA mechanism. C5a in the sample is bound to the capture antibody immobilized on the 96-well strip plate and then sandwiched with the biotinylated C5a antibody. After the addition of HRP-avidin and TMB substrate, the solution in the wells turns blue. The color reaction is stopped by adding the stop solution into the wells, and the color changes from blue to yellow. The color intensity is positively proportional to the C5a bound in the initial step. The C5a concentration can be calculated according to the standard curve. This kit is tested with high sensitivity and strong specificity.

C5a is a pro-inflammatory polypeptide that is generated when the complement cascade of serum proteins is activated in response to external surfaces like bacteria and tissue that has been injured by physical or chemical harm. C5a has potent biological actions that result in inflammatory consequences. C5a is a strong chemoattractant that plays a role in the recruitment of inflammatory cells like neutrophils, eosinophils, monocytes, and T lymphocytes, as well as the activation of phagocytic cells, the release of granule-based enzymes, and the generation of oxidants, all of which can contribute to innate immune functions or tissue damage. Increasing evidence suggests that C5a serves as a crucial link between innate and adaptive immune processes, potentially expanding C5a's role in inflammation. Overexpression or under-regulation of C5a is implicated in human and/or experimental models of inflammatory conditions, such as rheumatoid arthritis (RA), and osteoarthritis, and inflammatory bowel diseases.
